oil immersion

A technique used in light microscopy in which a special oil-immersion objective lens is employed to increase the resolving power of the microscope. A specimen mounted on a glass slide is covered by a drop of clear oil placed on the coverslip. The oil has the same refractive index as the glass lens. When the lens is immersed in the oil, the amount of light entering the objective increases (i.e. the effective aperture is increased). Such a technique is particularly useful at high magnifications. Oil is traditionally used but other liquids (e.g. sugar solution) may also be suitable.
