aceto-orcein

A stain used in the preparation of root-tip or anther squashes for chromosome examination. The material to be stained is placed in the aceto-orcein (which is acidified with hydrochloric acid) and heated at 60°C for 15 minutes. The material is then removed and mounted in acetic acid.
